Davidovich Fokina, ranked No 18, led 7-6 (4), 3-1 when Humbert, ranked No 24, pulled out at the St. Jakobshalle. in Basel on Saturday evening. He had called for a medical time out and a care on his back.
The Spaniard will play Brazilian Joao Fonseca on Sunday.
Alejandro Davidovich Fokina defeated Norwegian Casper Ruud, the No 4 seed (7-6 (1) ret.), American Jenson Brooksby (6-7 (6), 6-4, 7-5) and Italian Lorenzo Sonego (7-6 (2), 6-4) ahead of his victory.
Earlier in the tournament, the Frenchman won against American qualifier Reilly Opelka (7-6, 6-4), American Taylor Fritz, the top seed (6-3, 6-4) and American Sebastian Korda (6-3, 6-4).
Davidovitch Fokina and Humbert are supposed to meet again at the Rolex Paris Masters first round.
