American Iva Jovic, the No 8 seed, lost 7-6 (5), 6-2 to Romanian qualifier Gabriela Lee at the Fifth Third Charleston 125 2 on Thursday evening.
Lee, ranked No 306, will face American wildcard Lauren Davis next.
The Romanian won against Canadian lucky loser Kayla Cross (6-2, 6-1) in the previous round.
In the 32, Jovic, ranked No 207, defeated wildcard Usue Maitane Arconada (6-4, 6-4).
